Shire grew up in Brookline. She is the chef at Scampo at the Liberty Hotel (formerly Charles Street Jail) In 1986, she was the first female Executive Chef at the Four Seasons Hotels and Resorts in Beverly Hills. In 2001, she purchased Locke-Obers with Paul Licari, and, historically, became the first female chef. She has been chef at Bibo and Excelsior as well as Blue Sky in York Beach Maine. In 1992, she was recipient of James Beard Foundation Award America's Best Chef in the Northeast. In 1996, she was nominated as one of the 5 best chefs in the United States

Chef Shire's kitchens have been a training ground for some of the city's finest culinary talent, such as Jody Adams, Dante de Magistris, Gordon Hamersley, Amanda Lydon and Susan Regis.
